Effect of Medium on Regeneration and Growth of Bulblets from Culture of Bulb Scale Segments of hyacinthus orientalis in Vitro

Abstract

|
In vitro culture was applied to study the regeneration of bulblets on bulb scale segments of Hyacinthus orientalis cvs. Lady Derby, Mary, and Jan Bos. Bulblets with primary roots were obtained directly on a normal Heller´s medium. Root formation from bulblets was also delayed, as the explanting date was delayed. During the subcultures of separate bulblets, the bulbet growth was almost inhibited by treatments of 0.1 ㎎·L^(-1) or 1 ㎎·L^(-1) 6-benzylamino purine (BA), and an early tissue of cv. Mary turned brown. However root and bulblet growth of seperate bulblets without adventitious root was promoted on medium supplemented with 1 ㎎·L^(-1) IAA+0.1 ㎎·L^(-1) BA or 0.05 ㎎·L^(-1) NAA+0.1 ㎎·L^(-1) BA.
